Abstract | Protoplasts from cultured cells of Vicia hajastana and brome grass (Bromus inermis) were prepared by enzymatic digestion. Cell regeneration and division occurred and embryos were produced from the brome grass protoplasts. Studies were made on the influence of sucrose and calcium concentrations. Addition of 5mM CaCl₂ and I mM CaH₄(PO₄)₂ yield a higher percentage of dividing cells. The beneficial and adverse effects of sucrose was directly related to calcium concentration. Supplementing the medium with 20 mM NH₄NO₂ reduced cell division irrespective of the amount of calcium present. |
