Abstract	Endothelins are a group of 21-amino acid peptides secreted by endothelial cells, and expressed as three isoforms in the human genome (endothelin-1, -2 and -3) (6). Endothelin-1 (ET-1) has potent vasoconstrictor activity both in vitro and in vivo (8, 22). In addition to endothelium, presence and/or secretion of immunoreactive ET-1 has been shown in vascular smooth muscle cells, various glial cells, and neurons (5, 12, 15). Vasoactive actions of endothelins are mediated by two types of endothelin receptors characterized as ETA, with high affinity for ET-1 and low affinity for ET-3, predominantly localized on vascular smooth muscle cells, and nonselective ETB, mainly expressed on endothelial cells (7). Intracellular signaling events mediating vasoactive actions of endothelins depend on the type of receptors, tissue and species investigated (7, 14, 20), and comprise activation of phospholipase C (PLC), subsequent inositol triphosphate (IP3)-mediated mobilization of intracellular calcium, diacyl glycerol (DAG)-mediated stimulation of protein kinase C (PKC), activation of phospholipase A2 (PLA2) and arachidonic acid release, and influence on ionic transport across the membranes (7,16,20).
