Abstract | Analysis of algal biotoxins in testing and research laboratories has been moving steadily towards the use of LC-MS methods. This has been most challenging for paralytic shellfish toxins (PSTs) due to their polarity, the presence of multiple isomers and the complexity of sample matrices. Differential mobility spectrometry (DMS) allows for the separation of gas phase ions produced by electrospray ionization, at atmospheric pressure, prior to MS detection. The orthogonality of the DMS separation to LC and MS makes it suitable for use as a filter to remove chemical interferences in a multidimensional LC-DMS-MS configuration. This poster shows progress in implementing this technique for the analysis of PSTs, including a detailed study of the DMS separation of toxins and isomers with the goal of minimizing co-eluting matrix interference and separating toxin isomers from one another. The accurate metering of solvent vapor modifiers in the DMS carrier gas was particularly important for achieving sensitive and selective DMS separations. These studies also offered insight into the dissociation of labile PST ions during DMS separation. Optimized DMS conditions were then combined with hydrophilic interaction liquid chromatography (HILIC) and selected reaction monitoring (SRM) methods to give a highly selective HILIC-DMS-MS/MS method. Robust quantitative analysis using matrix matched calibration is demonstrated as well as the utility of this technique for eliminating matrix interference and increasing selectivity. |
