Abstract | 17 Strains of Rhizobium leguminosarum which varied in phenotypic expression of uptake hydrogenase (Hup) were analyzed for the presence of hup-specific DNA sequences using hybridization probes constructed from hup-specific DNA cloned from Rhizobium japonicum (PHVT100) and R. leguminosarum (PHVT109 and PHVT115). 2 Hup− strains and 3 strains with trace amounts of Hup activity showed no hybridization. 10 Hup⁺ strains hybridized with all 3 probes, indicating that hup-specific DNA sequences are conserved in these strains. 2 Hup⁺ strains did not hybridize with any of the probes. These strains may possess significantly different DNA sequences determining the Hup phenotype. |
