| Abstract | Embryoids were initiated from alfalfa mesophyll protoplasts and subsequently regenerated to plants. Leaves from fast growing plants under low light intensity were preferred for protoplast isolation. Complete release of mesophyll protoplasts from leaflets was achieved in an enzyme solution consisting of 0.5 % Onozuka cellulase, 0.5 % Rhozyme, 0.25 % pectinase, 300 mg/l calcium and a 0.5 osmalal within 5–6 hrs. at 23–25 °C.
Considerable variations in cell division were found in protoplasts produced from different plants. Protoplasts from younger leaves usually divided sooner than those from older leaves. In most cases the frequency of cell division was higher when the ratio of packed protoplast volume to medium volume was 0.1 % or lower. The ability of these protoplasts to form embryoids and regenerate into plants varied considerably from plant to plant within the same cultivar. |
