Résumé | Immunofluorescence light microscopy has made a significant impact on plant cell biology over the past decade. It is a powerful yet convenient tool, as it offers all the advantages of the conventional light microscope but with increased specificity and sensitivity. It has found application in numerous investigations of plant cells, particularly those related to the plant cytoskeleton (Lloyd 1987). Immunofluorescence light microscopy permits the rapid examination of the three-dimensional organization of microtubules (MTs) in fixed whole cells of plants. Immunofluorescence methods have recently been developed for sections of plastic-embedded plant material (Baskin et al. 1992), thus permitting examination of fluorescently labeled macromolecules in situ. |
