Résumé | A fast growing cell suspension culture of sorghum (Sorghum bicolor L. Moench) has been established. Out of 15 genetic strains, cv. GPR-168 responded most favorably to conditions for callus formation. The callus was friable and upon transfer to liquid medium produced cell suspension cultures. During 96 h of subculturing the cells showed mitotic indices of 1.0 to 6.5 %. Plant regeneration from callus derived from seedlings and immature embryos was achieved in two sorghum strains. Cytology of callus, cell suspension cultures, and regenerated plants indicated a strong prevalence of polyploidization.
The study also includes observations on a fast growing cell culture of corn. The culture had a doubling time of 48 h and during a 72 h period showed mitotic indices from 3.6 to 10.9 %. Protoplasts were readily isolated from the cell culture, but spontaneous fusion occurred extensively. On average 52 % of the isolated protoplasts were multinucleated. Attempts to eliminate the phenomenon were not successful. During subsequent culture some of the protoplasts regenerated cell walls and limited division occurred. |
