Résumé | Sustained division has been demonstrated in protoplasts from cultured cells and plant organs after reformation of the walls. Protoplasts are cultured by a liquid droplet method and on agar plates. The liquid droplet method facilitates gas exchange and feeding the newly formed colonies, which the plating techniques facilities continued observations of individual protoplasts. Using mixtures of sugars is superior to s single carbon source. Successful culture depends on nutritional and environmental factors. The optimum osmolality generally varies from 0.3 to 0.6. Cytokinesis is proceeded by cell wall regeneration. The newly formed wall may differ in structure from that present in cells formed on division. Cell regeneration and division has been observed in uninucleate as well as multinucleate homokaryons. Plants were grown from protoplasts of tobacco mesophyll, and from carrot and brome grass cells. The carrot and brome grass were produced by embryogenesis. |
