Abstract | A method was developed to determine the cell number of cell aggregates and embryos. The method was employed to study the rate of cell proliferation and frequency of embryo formation in a cell suspension culture of anise (Pimpinella anisum L.). The efficiency of embryogenesis was affected by the initial cell concentration (population density). In addition, a close relationship existed between the rate of cell division in cell aggregates and the efficiency of embryo-genesis. Maximum embryogenic efficiency and rate of cell division occurred in cultures with an initial population density of 10⁵ cells/ml. Cell proliferation in creased several-fold during the transition from the “heart-stage” to the “torpedo-stage”. This burst of cell division was preceded by a transient rise of the average cell volume. |
