Abstract | A particulate preparation of cyclic AMP phosphodiesterase from rat thymic lymphocytes exhibited two apparent Km's at 0.9×10−6M and 8.0×10−6M. The enzyme with the higher Km was stimulated by cyclic GMP by a mechanism involving an increase in the Vmax of the enzyme with no change in the Km. Cyclic GMP competitively inhibited the enzyme with the low apparent Km which had a Ki for cyclic GMP of 4×10−5M. The modulation of cyclic AMP phosphodiesterase activity by cyclic GMP in the control of cyclic AMP-mediated lymphocyte proliferation is discussed. |
