Abstract | Complete automation of precisely controlled cell culture microenvironment is herein presented on a centrifugal microfluidic platform with pneumatic active control interface (Powerblade) [1]. The proposed approach to cell culture and stimulation allows for humidity, CO2 and temperature control, analogous to traditional cell culture environments, whilst perfusing cells with the appropriate reagent buffers on-demand. The disposable passive cartridges were fabricated in standard thermoplastic polymers, which allows future use of scalable manufacturing methods suitable for industrial translation. Culture and stimulation of peripheral blood mononuclear cells (PBMCs) is demonstrated and cell activation is validated using ELISA. |
